Abstract

El sistema inmune innato supone la primera línea de defensa frente a patógenos. Para ello, células como los macrófagos disponen de múltiples receptores que les permiten distinguir lo propio de lo ajeno y montar una respuesta inflamatoria capaz de contener las infecciones y reparar el daño producido. Estos receptores pueden localizarse en la membrana celular, como los TLRs, entre los que destaca el TLR4 capaz de reconocer moléculas como el LPS. Pero también se conocen receptores intracelulares entre los que se encuentran miembros de la propia familia de TLRs y otros receptores como RIG que reconocen ácidos nucléicos, y además receptores de la familia NOD-like (NLRs) capaces de reconocer múltiples efectores. Interesantemente, algunos de estos receptores tienen una función muy distinta a todos los anteriores mencionados, ya que no desencadenan cascadas de señalización encaminadas a la activación de receptores nucleares y cámbios en la expresión génica celular, sino que producen complejos citosólicos multiprotéicos, conocidos como inflamasomas, encargados de proteolizar poderosas interleuquinas proinflamatorias, como es el caso de la Il-1b y IL-18.
