El déficit en la proteína similar a la mielina P0 tipo 2 causa hipoacusia prematura progresiva

Abstract

La proteína similar a la mielina P0 tipo 2 (MPZL2) media interacciones célula-célula en diversos tejidos como riñón o pulmón. En familias consanguíneas se han identificado variantes de MPLZ2 asociadas a hipoacusia temprana, moderada, simétrica y progresiva, con mayor afectación de frecuencias altas. Para profundizar en las funciones MPLZ2 en la cóclea y comprender las bases moleculares de la hipoacusia causada por su déficit, hemos realizado el estudio comparado de ratones Mplz2-/- y de sus controles silvestres. Nuestros resultados indican que MPZL2 se localiza en las células de Deiters, células ciliadas y células basales de la estría vascular. Los potenciales evocados auditivos realizados a las 4, 8 y 12 semanas de edad, indican que los ratones nulos, pero no los controles, presentan hipoacusia desde las 4 semanas de edad, siendo ésta bilateral y progresiva, y afectando principalmente a las frecuencias altas. La deficiencia en MPLZ2 causa desorganización de la citoarquitectura y degeneración del órgano de Corti, y pérdida de neuronas del ganglio espiral, siendo más pronunciadas en la base de la cóclea. En conclusión, las mutaciones deletéreas en Mplz2/MPLZ2 producen pérdida auditiva temprana en el hombre y en el ratón. El déficit en esta proteína afecta potencialmente a la adhesión celular en el epitelio ótico causando pérdida de integridad estructural y degeneración progresiva del órgano de Corti y, secundariamente, del ganglio espiral.
