Abstract

[ES]: La transcripción es un proceso esencial, que en eucariotas es llevado a cabo por tres RNA polimerasas (RNAPI, II y III). La RNAPII se encarga principalmente de transcribir todos los genes que codifican para proteínas (mRNA) y algunos RNAs no codificantes pequeños. El proceso de transcripción se encuentra finamente regulado en todas sus etapas (pre-iniciación, iniciación, elongación y terminación), por numerosos factores. Además, la biogénesis de los mRNA se encuentra regulada y coordinada con otros procesos nucleares, como son la modificación de histonas, la remodelación de la cromatina, la reparación del DNA, la recombinación y la replicación. Sin embargo, por el momento, ningún estudio ha revelado una relación funcional entre los complejos implicados en el mantenimiento de los telómeros y la maquinaria de transcripción por la RNAPII. En este TFG, hemos investigado el papel del complejo telomérico CST en la regulación de la transcripción por la RNAPII en Saccharomyces cerevisiae. Nuestros resultados indican que el complejo CST, a través de una de sus subunidades, Ten1, tiene un papel en la elongación de la transcripción, regulando la asociación del complejo de elongación Spt4/Spt5 a la cromatina. Así mismo, parece ser que Ten1 podría influir sobre los niveles de fosforilación de la RNAPII. Dicha fosforilación es clave en la regulación de la transcripción. Por lo tanto, en conjunto, nuestros resultados, sugieren un papel importante del complejo telomérico CST en la transcripción.

[EN]: The transcription is an essential process which in eukaryotes is carried out by three RNA polymerases (RNAPI, II y III). The RNAPII transcribes all genes that code for proteins (mRNA) and some small non-coding RNAs. The transcription is highly regulated by large factors in its all stages (pre-initiation, initiation, elongation and termination). In addition, the biogenesis of mRNA is regulated whit others nuclear process such as histone modification, chromatin remodeling, DNA repairing, recombination and replication. However, for the moment, any studies have shown a functional relationship between the complexes involved in the maintenance of telomeres and the transcription by RNPII. In this TFG, we have investigated the role of telomeric complex CST in the regulation of transcription by the RNAPII in Saccharomyces cerevisiae. Our results show that the CST complex, through one of its subunits called Ten1, has a role in the elongation of transcription. In particular, Ten1 seems to regulate association of the elongation complex Spt5/Spt4 to the chromatin. Also, Ten1 could affect phosphorylation levels of RNAPII that is an essential process in the regulation of transcription. Therefore, our results suggest an important role for the CST complex in transcription.
