[EN] The TFG has been performed at the IBFG (CSIC-USAL) within the transcriptional regulation group. This group aims to elucidate some of the mechanisms involved in the regulation of transcription by RNA polymerase II (RNAPII), and RNAPI in eukaryotes. For that purpose the yeast Saccharomyces cerevisiae yeast is used as model organisms. Previous studies carry out in the laboratory showed that Sub1, likely through its C-terminal region, interacts with RNAP II stalk domain. In addition, studies performed by others showed that Sub1 has a role during RNA polymerase III (RNAPIII) transcription. Regarding RNAPI, there are controversial studies involving Sub1 in RNA polymerase I (RNAPI). Therefore, due to the structural homology between all the three RNA polymerases, it is possible that Sub1 associates with all of them through their respective stalk domains. On the other hand, Sub1 has been involved in transcription elongation via the interaction with the elongation complex Spt4/5, though in this case it is unknown which region of Sub1 interacts with this complex. Our hypothesis is that it may occur through the C-terminal region of Sub1, because preliminary data obtained in the laboratory indicate that this region influences transcription elongation. Therefore, the objectives of this TFG are: first, to study if the transctiptional factor Sub1 interacts with RNA polymerase I (RNAPI); and second to investigate if the carboxi-terminal region of Sub1 may be involved in the interaction with Spt4/5 during transcription elongation. To achieve this objective, we employed genetic and biochemical techniques, such as co-immunoprecipitation and Western-blot analysis. Unfortunately, no positive results were obtained, mostly due to technical problems.
[ES] Este TFG se ha realizado en el IBFG (CSIC-USAL) dentro del grupo de regulación transcripcional. Este grupo tiene como objetivo dilucidar algunos de los mecanismos involucrados en la regulación de la transcripción por la RNA polimerasa II (RNAPII) y RNAPI en eucariotas. Para ello, se utiliza como organismo modelo la levadura Saccharomyces cerevisiae de levadura. Estudios anteriores realizados en el laboratorio mostraron que Sub1 interactúa con el dominio del tallo (stalk) de la RNAP II, probablemente a través de su región C-terminal. Además, otras investigaciones realizadas en diferentes grupos demostraron que Sub1 tiene un papel durante la transcripción de la RNA polimerasa III (RNAPIII). Con respecto a RNAPI, existen estudios controvertidos que involucran a Sub1 en la RNA polimerasa I (RNAPI). Por lo tanto, debido a la homología estructural entre las tres RNA polimerasas, es posible que Sub1 se asocie con todos ellos a través de sus respectivos dominios de tallo. Por otro lado, Sub1 está involucrado en el proceso de elongación de la transcripción a través de la interacción con el complejo de elongación Spt4 / 5, aunque en este caso se desconoce qué región de Sub1 interactúa con este complejo. Nuestra hipótesis es que puede ocurrir a través de la región C-terminal de Sub1, porque los datos preliminares obtenidos en el laboratorio indican que esta región influye en la elongación de la transcripción. Por lo tanto, los objetivos de este TFG son: primero, estudiar si el factor de transcricpción Sub1 interactúa con la RNA polimerasa I (RNAPI); y segundo, investigar si la región carboxilo-terminal de Sub1 puede estar involucrada en la interacción con Spt4 / 5 durante la elongación de la transcripción
