La biosíntesis del glucano y su función en la citocinesis y en el crecimiento polarizado

Abstract

La pared celular es una estructura esencial en las células fúngicas que se encuentra ausente en las células de animales. Es una estructura rígida, externa a la membrana plasmática, que proporciona a la célula protección mecánica y resistencia a la presión osmótica interna. A pesar de su rigidez, la pared celular es capaz de adaptarse a los cambios morfológicos que se producen durante el ciclo de vida de la célula fúngica. Así, durante la citocinesis se produce la biosíntesis de un septo de división simultánea a la contracción de un anillo de actomiosina. La levadura de fisión Schizosaccharomyces pombe es un hongo unicelular con forma bacilar que crece por extensión de los extremos y se divide por fisión en la zona media, generando dos células hijas del mismo tamaño. S. pombe es un organismo modelo para el estudio de la citocinesis y el crecimiento polarizado porque su morfología, tamaño y división son extremadamente reproducibles. La pared celular de S. pombe posee dos tipos de glucanos como componentes estructurales mayoritarios y esenciales: ß-(1,3)-D-glucano con algunas ramas ß-(1,6) y ¿-(1,3)-D-glucano. Bgs4 y Ags1 son las enzimas responsables de la síntesis de estos polímeros. También hay una pequeña cantidad de ß-(1,3)-D-glucano lineal localizado en el septo primario y en las áreas de crecimiento. Bgs1 es la enzima responsable de la síntesis de este ß-(1,3)-glucano lineal que es esencial para la formación del septo y para la viabilidad celular. En esta ponencia se presentan resultados recientes que profundizan en el conocimiento del proceso de septación de la levadura de fisión describiendo la colaboración de Bgs1 con proteínas del anillo de actomiosina, como la paxilina y la fosfatasa calcineurina, en la formación del surco de división. Además, presentaremos resultados preliminares sobre la función de Bgs1 y el posible papel del glucano lineal en el establecimiento del crecimiento polarizado.
