Abstract

Previamente hemos descrito que el flavonoide sintético 3,4¿-dimetoxi-3¿-hidroxi-flavona presenta una potente actividad antiproliferativa en varias líneas tumorales hematopoyéticas, incluídas aquellas que expresan niveles elevados de las proteínas anti-apoptóticas Bcl-2 y Bcl-xL. En este estudio hemos evaluado sus efectos sobre el ciclo celular, los cambios sobre la expresión y la fosforilación de las proteínas reguladoras del ciclo y sobre la polimerización de tubulina y la red de microtúbulos. Las líneas celulares utilizadas en este estudio fueron HL-60, U937 y Molt-3. La distribución de las fases del ciclo celular se determinó por citometría de flujo de células teñidas con yoduro de propidio. Los cambios de las proteínas reguladoras del ciclo celular se determinaron por transferencia Western. El efecto sobre la polimerización de tubulina se evaluó cinéticamente utilizando un lector de microplacas. Los experimentos para determinar el efecto sobre los microtúbulos se realizaron por inmunofluorescencia. Los resultados ponen de manifiesto que el efecto de esta flavona sintética sobre la inhibición de la proliferación de las células leucémicas es debido a una parada del ciclo celular en la fase G2-M, que fue acompañada por un aumento de la fracción de células hipodiploides, y a la inhibición de la polimerización de tubulina. Esto estuvo asociado con una inducción y fosforilación de la ciclina B1, una acumulación del inhibidor del ciclo celular p21Cip1 y la activación de la proteína quinasa dependiente de ciclina Cdk1 vía desfosforilación de fosfoT14-Y15.
