[ES] La presente invención proporciona un procedimiento para producir compuestos esteroideos 11α hidroxilados o derivados de los mismos por fermentación de esteroles naturales o por biotransformación de distintas sintonas esteroideas con bacterias recombinantes, preferiblemente de Mycobacterium smegmatis o de Corynebacterium glutamicum. Dichas bacterias recombinantes portan un plásmido que comprende un operón sintético que contiene los genes cyp509C12 y roCPR1 de Rhizopus oryzae, que codifican el citocromo CYP509C12 con actividad 11α-hidroxilasa y la citocromo reductasa (RoCPR1) necesaria para la actividad del citocromo. Mediante el método descrito en la presente invención se pueden producir, aunque sin limitarnos, 11α OH-ADD o 11α-OH-AD a partir de esteroles naturales como colesterol o fitoesteroles, o 1α-OH-PROG, 11α-OH-DOC, 11α-OH-TEST, 11α-OH-DHEA, 11α-OH-AD y/o 11α-OH-ADD a partir de sus correspondientes sintonas no hidroxiladas.
[EN] The present invention provides a method for producing 11α-hydroxylated steroid compounds or derivatives thereof by fermentation of natural sterols or by biotransformation of various steroidal synthons with recombinant bacteria, preferably Mycobacterium smegmatis or Corynebacterium glutamicum. Said recombinant bacteria carry a plasmid comprising a synthetic operon containing the genes cyp509C12 and roCPR1 from Rhizopus oryzae, which code for cytochrome CYP509C12 with 11α-hydroxylase activity, and the cytochrome reductase (RoCPR1) needed for activity of the cytochrome. Using the method described in the present invention, 11α OH-ADD or 11α-OH-AD can be produced from natural sterols such as cholesterol or phytosterols, and 1α-OH-PROG, 11α-OH-DOC, 11α-OH-TEST, 11α-OH-DHEA, 11α-OH-AD and/or 11α-OH-ADD can be produced from the corresponding non-hydroxylated synthons, amongst others.
