Refuerzo del reconocimiento molecular

Abstract

La cooperatividad es uno de los fenómenos más importantes en biología molecular, ya que se encarga tanto de regular la afinidad de un sustrato por un receptor, como de las propiedades que emergen de los complejos que no están presentes en sus componentes individuales. Tradicionalmente, la cooperatividad ha estado relacionada con la multivalencia y con el alosterismo. Sin embargo, Williams y colaboradores han propuesto una interpretación ligeramente diferente y más general: la cooperatividad positiva proviene del refuerzo mutuo entre dos o más interacciones no covalentes, y como resultado, la energía libre global es mayor que la esperada de la suma de las contribuciones individuales. De acuerdo a esta interpretación parte de la fuerza impulsora del reconocimiento proviene del fortalecimiento de las interacciones existentes en el propio receptor, lo cual esta correlacionado con una compresión del complejo. A este tipo de cooperatividad se le ha denominado refuerzo del reconocimiento molecular, y es considerada como una de las grandes responsables de la alta afinidad de los receptores bióticos. Sin embargo, ha sido escasamente descrita en receptores sintéticos. Nuestro grupo de investigación ha desarrollado nuevos receptores quirales de cationes como modelos moleculares sencillos para el estudio y la comprensión de las interacciones no covalentes y de la cooperatividad existente entre las mismas.
