Modificación selectiva de péptidos: conversión de residuos >modulables> en aminoácidos no naturales de alta pureza óptica

Abstract

La modificación de péptidos es un área de gran interés tanto en química médica(obtención de péptidos bioactivos o componentes de fármacos) como en química sintética (obtención de péptidos catalíticos, foldámeros, etc). En el proceso tradicional para obtener análogos de péptidos, cada compuesto se sintetiza de novo, introduciendo una modificación respecto al péptido de referencia.Este procedimiento implica un gasto considerable de tiempo y materiales; por ello, una estrategia alternativa (site‐selective modification) consiste en manipular selectivamente determinados aminoácidos, sin afectar al resto de los residuos.1b‐e De esta forma, a partir de un único péptido (o de unos pocos) se pueden obtener bibliotecas de péptidos modificados, y establecer rápidamente relaciones estructura‐actividad. Pese a su interés, esta estrategia supone un reto difícil, debido a la reactividad similar de los distintos aminoácidos, especialmente si hay varios residuos modificables del mismo tipo.Aquí se detallará el desarrollo de nuevos amino ácidos “modulables”, y su conversión en derivados ópticamente puros de aminoácidos no naturales.
