Síntesis de 4-Amino-alfa-cetoácidos y sus Derivados Peptídicos a partir de alfa-Aminoácidos y Péptidos de pequeño tamaño

Abstract

Muchos -amino--cetoácidos (y sus derivados éster o amida) forman parte de potentes inhibidores de proteasas (trombina, calpaina, catepsinas, proteasa de HIV) que presentan actividad antitrombótica, antitumoral y antiviral, entre otras.1 Por ello, los derivados de -peptidil--cetoácidos están siendo muy estudiados. En contraste, se ha hecho muy poco trabajo para determinar la actividad biológica de -amino--cetoácidos y sus derivados peptídicos. En esta comunicación se describe un método para generar fácilmente bibliotecas de estos compuestos que permitan realizar estudios estructura-actividad. La metodología implica la -fragmentación radicalaria oxidativa de aminoácidos comerciales o pequeños péptidos, seguida de tratamiento con 2-(trimetilsililoxi)-acrilato de metilo en presencia de un ácido de Lewis. Así, se accede de forma rápida y eficiente a las estructuras de interés, utilizables como bloques de construcción de cadenas peptídicas más complejas.
