Modificación Selectiva de Péptidos. Conversión de Unidades de Serina o Treonina en Deshidroaminoácidos

Abstract

Los deshidroaminoácidos se encuentran en numerosos péptidos bioactivos. La introducción de deshidroaminoácidos en análogos sintéticos de péptidos bioactivos es una práctica de gran interés, pues puede mejorar la resistencia de éstos a la degradación enzimática, y permite una mejor modulación de sus propiedades. En nuestro grupo hemos desarrollado una metodología que permite transformar selectivamente residuos de serina o treonina de péptidos sencillos en unidades de deshidroaminoácidos sustituidos. El proceso se inicia con una -fragmentación radicalaria oxidativafosforilación del péptido 1, para obtener los aminofosfonatos 2. Éstos reaccionan con diferentes aldehídos mediante una reacción de HornerWadsworth-Emmons, dando lugar a péptidos con residuos de deshidroaminoácidos 3, con buenos rendimientos y alta estereoselectividad, evitando los largos y costosos métodos de modificación convencionales.
