Evaluación Antileucémica de 17 Derivados Sintéticos de Flavonoides en Células de Leucemia Humana

Abstract

Los flavonoides son compuestos polifenólicos naturales que contienen una estructura común de fenilbenzo-¿-pirona y desempeñan un amplio espectro de actividades biológicas. Las propiedades antitumorales de estos compuestos están mediadas por diferentes tipos de parada del ciclo celular y la inducción de apoptosis, una forma activa de muerte celular que desempeña un papel esencial en el desarrollo y la supervivencia y es una respuesta importante a la mayoría de los agentes quimioterapéuticos. En este estudio hemos determinado la actividad antileucémica potencial de 17 compuestos derivados de la fenilbenzopirona en tres líneas celulares de leucemia humana. De los compuestos ensayados, la 3,4¿-dimetoxi-3¿-hidroxi-flavona fue la más potente, ya que presentó las concentraciones inhibidoras 50% (IC50) más bajas en todas las líneas ensayadas. Los valores de IC50 fueron 0,8 ± 0,1 ¿M, 2,3 ± 0,3 ¿M y 5,4 ± 2,4 ¿M en HL-60, Molt-3 y U937, respectivamente. Los ensayos biológicos utilizados incluyeron la evaluación de citotoxicidad a través del ensayo de actividad metabólica utilizando el reactivo MTT, la microscopía de fluorescencia después de la tinción con el fluorocromotrihidrocloruro de bisbenzimida, la fragmentación del ADN, la microscopía electrónica de transmisión y la citometría de flujo. Este compuesto induce apoptosis, promueve la hidrólisis de múltiples caspasas (las procaspasas 3, 6, 7, 8 y 9) y la activación de la vía de señalización de las proteínas quinasas activadas por mitógenos. Además, este flavonoide es citotóxico para las células de leucemia que expresan niveles elevados de las proteínas anti-apoptóticas Bcl-2 y Bcl-xL, frecuentemente asociadas a fenómenos de resistencia.
