Abstract

[ES] Los grandes ensamblados macromoleculares presentan unas propiedades mecánicas extraordinarias las cuales los hacen perfectos para su uso con fines muy distintos dentro de la nanotecnología y la biomedicina. El virus diminuto del ratón y la partícula vault son dos ejemplos de ensamblados macromoleculares que podrían usarse con estos fines. La determinación estructural de la cápside del mutante N170A de MVM, el estudio de sus propiedades mecánicas y la comparación con el mutante L172W ha permitido entender mejor la base estructural que hay detrás de la rigidificación de la cápside causada por estos dos tipos de mutaciones que afectan al ciclo infectivo del virus. Por otro lado, la caracterización de las propiedades mecánicas de las vaults y el estudio de los distintos mutantes de MVP por diversas técnicas, proporciona nueva información sobre el comportamiento dinámico de la particula, así como sobre su mecanismo de apertura, que podría ayudar a entender el papel desempeñado por esta partícula en la célula. Por último y dentro del estudio de esta particula, se ha llevado a cabo la caracterización estructural por cryo-EM de la vault en complejo con la proteína TEP1, resultando en la obtención de la primera estructura de esta proteína a baja resolución. El estudio de la misma ha mostrado que esta proteína presenta una alta heterogeneidad estructural en solución.

[EN] The large macromolecular assemblies have extraordinary mechanical properties make them perfect to use them in nanotechnology and biomedicine. The minute virus of mice (MVM) and the vault particle are two examples of macromolecular assemblies that could be used for these purposes. The structural determination of the capsid of the MVM N170A mutant, the study of its mechanical properties and the comparison with the mutant L172W has allowed a better understanding of the structural basis behind the rigidification of the capsid caused by these two types of mutations that affect to the infective cycle of the virus. On the other hand, the characterization of the mechanical properties of the vaults and the study of the different MVP mutants by different techniques, provides new information about the dynamic behavior of the particle, as well as its mechanism of opening, which could help to understand the role played by this particle in the cell. Finally, within the study of this particle, the structural characterization by cryo-EM of the vault in complex with the TEP1 protein has been carried out, obtaining the first structure of this protein at low resolution. The study has shown that this protein has a high structural heterogeneity in solution.
