[Introduction]: The purpose was to evaluate the effects of by-products of Ceratonia siliqua L. on TG accumulation in 3T3-L1 mature adipocytes and inflammatory response amelioration in RAW 264.7 macrophages which has not been described. [Material and Methods]: Methanolic extractions from germ, seed-peel and pod were performed. 3T3-L1 mature adipocytes were treated during 24h with extracts at 0.5, 0.1 and 0.05mg/mL dose, and TG content was quantified. Anti-inflammatory activity was measured through determination of NO, TNF-α and PGD2 in LPS-stimulated RAW 264.7 macrophages. Cell viability was determined. [Results]: Twenty-five phenolic compounds, belonging to hydroxybenzoic acids, gallotannins, flavones and flavonols were identified. No cytotoxic effect was observed for any dose of extracts in adipocytes. Seed-peel and pod extract promoted TG reduction in adipocytes at a dose of 0.1 mg/mL. All extracts showed no cytotoxic effect in LPS-stimulated macrophages at a dose of 0.05 mg/mL. At this concentration, pod extracts produced a higher decrease of NO production in macrophages than seed-peel and germ extracts. PGD2 production was also reduced in macrophages treated with 0.05 mg/mL of germ and pod extracts although no effect was observed on TNFα. [Conclusion]: Seed-peel and pod extracts of Ceratonia siliqua L showed moderate anti-obesity and anti-inflammatory properties.
