Estudio del apareamiento entre cromosomas de distintas especies de cebada en el fondo genético del trigo harinero

Abstract

La hibridación in situ es una herramienta muy útil en programas de mejora genética porque permite identificar introgresiones de cromosomas completos, translocaciones, así como fragmentos cromosómicos de especies de interés en el fondo genético de otra especie. En nuestro caso, la hibridación in situ con ADN genómico (GISH) nos ha permitido estudiar durante la mciosis la asociación entre cromosomas de distintas especies de cebada (Hordeum chilense y H ordeum vulgare) en el fondo genético del trigo harinero ( Triticum aestivum cv. Chinese Spring). En concreto, hemos construido mediante cruzamientos genéticos, líneas de trigo que contienen un cromosoma de cada una de las la cebadas silvestre y cultivada (H. chilense y H. vulgare)pertenecientes tanto al mismo grupo de homeología como a distintos, y hemos estudiado mediante GISH el apareamiento cromosómico durante la meiosis en la Fl. Resultados preliminares muestran que en estadías tempranos de la meiosis (paquiteno) se produce apareamiento y asociación entre los cromosomas de ambas especies de cebada cuando pertenecen al mismo grupo tk homeología. Sin embargo, sólo ('n un muy bajo por('cutajc de células se observa apareamiento entre los cromosomas de ambas especies cuando éstos pertenecen a grupos de homeología diferentes. Además, las observaciones del apareamiento cromosómico en estadíos posteriores de la meiosis ( diploteno, diacinesis y metafase I) sugieren que aunque los cromosomas de las distintas especies de cebada pueden asociarse en paquiteno en el fondo genético del trigo, no se produce recombinación entre ambos, pues en ningún caso detectamos bivalentcs. Los resultados obtenidos hasta el momento así como sus implicaciones biológicas se discuten en este trabajo.
