Abstract

[ES]: En esta Tesis Doctoral se estudia el papel de BAMBI (BMP and activin membrane bound inhibitor), una proteína inhibidora de la señalización de TGFβ, BMPs y activinas, en la regulación de la respuesta inmune humoral. BAMBI se expresa a nivel basal en los linfocitos B, aumentando su expresión tras activación. Mediante el uso de ratones mutantes (BAMBI-KO), se observa que la deficiencia en BAMBI no afecta al desarrollo de estas células en la médula ósea, aunque en el bazo se observa un aumento en las subpoblaciones B T2, B memoria y B-1 no convencionales. Los ratones BAMBI-KO presentan un aumento en la producción de anticuerpos frente a antígenos T-independientes y T-dependientes en diferentes contextos de inmunización, en correlación con un aumento en la expansión de las células implicadas en la respuesta humoral (células T foliculares y células B del centro germinal). Este fenómeno es debido a un defecto intrínseco en las células B, las cuales tienen una expresión incrementada de moléculas de activación e interacción T-B y una mayor respuesta proliferativa. El aumento en la respuesta humoral observado en los ratones BAMBI-KO es dependiente de la señalización de TGFβ a través de TAK1. Este aumento de la respuesta humoral en protocolos de inmunización no se asocia con el desarrollo espontáneo de patologías autoinmunes de base humoral.

[EN]: We have studied the potential role of BAMBI (BMP and activin membrane bound inhibitor), an inhibitor of TGFβ, BMPs and activin signaling, in the regulation of the humoral immune responses. BAMBI is expressed at low levels in B lymphocytes at steady state and its expression increases after activation. Using BAMBI knock-out (KO) mice, we have observed that BAMBI deficiency does not affect the development of B cells in the bone marrow, although it promotes an increase in T2 B, memory B, and B-1 subpopulations in the spleen. BAMBI-KO mice show increased production of antibodies against T-independent and T-dependent antigens in different immunization contexts, in correlation with the expansion of all lymphoid populations involved in humoral response (follicular T cells and germinal center B cells). This phenomenon is secondary to an intrinsic B-cell defect, which have a higher expression of surface molecules involved in T-B cell interaction and activation and greater proliferative responses in vitro. The augmented humoral response observed in BAMBI-KO mice is selectively due to an increase in TGFβ signaling through TAK1. The enhanced humoral responses observed in BAMBI-KO mice after vaccination are not accompanied by the spontaneous development of autoantibody-dependent autoimmune pathologies.
