Clasificación de plásmidos por relaxasas

Abstract

Los plásmidos son elementos genéticos extracromosómicos implicados en la aparición de microorganismos multi-resistentes, dada su capacidad para transferir horizontalmente genes de resistencia a antibióticos. Por tanto, es importante contar con un método de clasificación de plásmidos eficaz que facilite su estudio epidemiológico. Los métodos tradicionales de clasificación, basados en los mecanismos de replicación del plásmido (PCR-Based Replicon Typing; PBRT), presentan problemas diversos. En este trabajo, definimos el método de clasificación de relaxasas por cebadores degenerados (Degenerated-Primer MOB-Typing; DPMT) y lo comparamos con PBRT. Para ello, re-analizamos seis colecciones de aislados bacterianos, previamente clasificadas PBRT. DPMT tiene una exhaustividad y especificidad muy alta. Clasifica un porcentaje de aislados mucho mayor que PBRT (94,4% frente a 80,3%). Pero DPMT deja sin clasificar algunos aislados sí clasificados por PBRT, por lo que son métodos complementarios. También se secuenciaron 18 plásmidos de entre los detectados en las seis colecciones analizadas. Se hizo un análisis de cada uno de ellos, comparándolos con la de las secuencias más próximas filogenéticamente.

El presente trabajo fue realizado en las instalaciones del Departamento de Biología Molecular de la Universidad de Cantabria, entre los años 2006 y 2011. Durante este tiempo, el autor de este trabajo estuvo contratado como Técnico de Apoyo a la Investigación, gracias a un proyecto financiado por IDICAN (2007-2010) y, posteriormente, por la propia Universidad de Cantabria (septiembre-diciembre, 2011).
