Análisis de los genomas de dos cepas de Pseudomonas spp. endófitas naturales de raíces de olivo (Olea europaea L.) y eficaces agentes de control biológico de la Verticilosis

Abstract

Pseudomonas fluorescens PICF7 y Pseudomonas putida PICP2 son dos cepas efectivas en el control biológico de la Verticilosis (Verticillium dahliae Kleb.), una de las enfermedades más devastadoras que afectan al olivar. Además, las cepas PICF7 y PICP2 pueden colonizar endofíticamente las raíces de olivo, estableciéndose de forma estable en los espacios intercelulares del córtex radical, incluso de forma simultánea. Sin embargo, nada se sabe de los determinantes bacterianos implicados en su actividad de biocontrol, ni de aquellos involucrados en el desarrollo de un estilo de vida endofítico. En este estudio hemos secuenciado y anotado los genomas completos de las cepas PICF7 (1 ‘contig’, 6,14 Mpb) y PICP2 (63 ‘contigs’, 5,60 Mpb). Se ha efectuado un análisis bioinformático comparativo dirigido a identificar genes que puedan estar implicados en (1) control biológico, (2) interacción planta-bacteria y (3) endofitismo. Este análisis ha revelado que, entre otros determinantes ampliamente estudiados como involucrados en biocontrol y/o interacción planta-bacteria, el genoma de la cepa PICF7 potencialmente codifica para dos sistemas de secreción T6SS y otro T3SS, los efectores T3SS avrE1 y hopB1, los sideróforos pioverdina y pioquelina, factores de adhesión y MDR, diversos enzimas degradadores de la pared celular vegetal (proteasa, celulasa y lipasa), así como sistemas de destoxificación tales como CopA, KatB, Dps y Cbb. Por el contrario, el genoma de PICP2 mostró, entre otras diferencias, ausencia de regiones codificantes para el sistema de tipo T3SS. Algunos de estos fenotipos se han comprobado mediante los correspondientes ensayos bioquímicos y microbiológicos. La disponibilidad de ambos genomas puede ayudar a comprender tanto la actividad de biocontrol como el modo de vida endófito que ambas cepas son capaces de desarrollar.
