Abstract

[ES]La tesis se ha centrado en la caracterizaci?n del fenotipo celular en ausencia de Cdh1, prestando especial atenci?n a esclarecer el papel de APC/C-Cdh1 en la prevenci?n de la inestabilidad gen?mica y del estr?s replicativo en modelos murinos. Se ha comprobado que la p?rdida de Cdh1 conduce a una replicaci?n ineficiente del DNA, provocando una alteraci?n de par?metros replicativos, tales como la velocidad de progresi?n de las horquillas o la frecuencia de activaci?n de or?genes. Esta situaci?n desencadena una activaci?n de la ruta de respuesta a da?o y una mayor acumulaci?n de roturas en el DNA de las c?lulas que carecen de Cdh1, poniendo en riesgo la estabilidad gen?mica. Como mecanismo responsable de dichos defectos replicativos en ausencia de Cdh1 se ha postulado la combinaci?n del exceso de actividad Cdk y las alteraciones en la homeostasis de los nucle?tidos, generando una limitaci?n de la concentraci?n de dNTPs a nivel de las horquillas.
