Infección experimental con mycobacterium avium subsp Paratuberculosis en cabritos previamente sensibilizados con Mycobacterium avium subsp hominisuis y corynebacterium pseudotuberculosis

Abstract

Una de las hipótesis que podrían explicar la diferente respuesta de los rumiantes a la infección por Mycobacterium avium subsp paratuberculosis (Map), incluyendo la mayor resistencia que ofrecen los animales adultos, es que su sensibilización previa con otras bacterias relacionadas modularía la respuesta inmunitaria e inflamatoria de forma que la evolución de las lesiones sea diferente. Se ha desarrollado una infección experimental en 23 cabritos que, a los 1,5 meses de edad fueron inoculados por vía oral con 5x103 ufc de Mycobacteriumn avium subsp hominissuis -Mah- (7 animales) o Corynebacterium pseudotuberculosis -Cp- (7 cabritos): en un total de 10 tomas administradas cada 3 días. A los 2 meses del Inicio, 5 cabritos de cada Grupo sensibilizado y otros 5 que no habían sido tratados, se infectaron por vía oral con 1,8x1010 ufc de la cepa 764 de Map. Se valoró la respuesta inmunitaria periférica celular (IFN-y) y humoral (ELISA indirecto) frente a Map, y a los 5 meses post-infección con Map se sacrificaron todos los anima!es. Se llevo a cabo el estudio histológico de distintas muestras de intestino y nódulos linfáticos, donde se valoró la presencia de lesiones granulomatosas y su localización así como la cuantificación mediante el recuento de granulomas. No se observaron diferencias en la producción de IFN-y y anticuerpos entre los tres grupos infectados con Map. En los cabritos infectados sólo con Map aparecieron lesiones granulomatosas en un mayor número de localizaciones intestinales, incluyendo zonas de yeyuno sin tejido linfoide, mientras que en los sensibilizados con Mah y Cp las lesiones estaban restringidas al tejido linfoide intestinal. Sin embargo, el número e intensidad de las lesiones fueron significativamente mayores en el grupo tratado con Cp+Map, que en el infectado con Map. El menor número se encontró en el grupo Mah+Map. En las condiciones de este estudio, la sensibilización previa con Mah y Cp influye en el desarrollo de lesiones en las fases tempranas de la infección por Map.
