The Passerini reaction is a pivotal isocyanide-based multicomponent reaction together with Ugi reaction and it provides easy access to α-acyloxy carboxamides in a one-pot synthesis involving an aldehydes, isonitriles, and a carboxylic acid. In this chapter, some pivotal examples will be remarked covering different aspects of this reaction. Interesting targets synthesized following this MCR as a key synthetic step will be also disclosed. Additionally, some interesting environmentally-benign protocols have been remarked.
