Papel del IGF-I/Ras en la diferenciacion de los adipocitos marrones fetales: Un nuevo mecanismo termogenico

Abstract

[ES]: Los adipocitos marrones fetales de rata son un modelo de células mesenquimales no fibroblásticas que presentan un alto número de receptores de alta afinidad para el IGF-I aumentando la proliferación celular, al mismo tiempo que se diferencian adipogenicamente (lo que conduce a un fenotipo de gotas de grasa multicolores)n y termogénicamente (asociada a la expresión de la proteína desacoplante). Además, hemos detectado la expresión endógena de IGF-I en estas células, lo que sugiere que el IGF-I puede ser una de las señales fisiológicas que están impulsando el desarrollo del tejido adiposo marrón in vivo durante los últimos días de vida fetal. En este sentido, el IGF-I aumenta la tasa de transcripción del gen de la UP y produce la acumulación de su mensajero, aumentando también la vida media del mismo. Esto conduce a un gran aumento en los niveles de proteína desacoplante detectables inmunológicamente. Igualmente, transfecciones transitorias de los adipocitos marrones fetales con el promotor completo del gen de la UP dirigiendo la expresión del gen bacteriano CAT demuestran que el IGF-I transactiva el promotor de la UP. Así mismo, el promotor de la UP se transactiva en ausencia de IGF-I cuando se cotransfectan las células con construcciones del Ras transformante, mientras que la cotransfección de Ras dominante negativo en presencia de IGF-I anula el efecto transactivador de IGF-I sobre el promotor de la UP. Estos resultados proponen un nuevo mecanismo termogénico mediado por IGF-I/ras en los adipocitos marrones fetales de rata.

[EN]: Foetal brown adipocyte primary cultures undergo proliferation in the presence of IGF-I and also induce the adipogenic-program related to lipid synthesis and the thermogenic-program associated with the expression of the uncoupling protein. Moreover, we have recently described an endogenous expression of IGF-I mRNA in brown adipose tissue at day 20 of foetal development and also an increase in the IGF-I receptor mRNA content at the end of the foetal life, which runs parallel to the onset of late tissue differentiation. Thus, IGF-I, in an autocrine/paracrine fashion may be a major signal involved in brown adipose tissue development prior to birth.
