Entouleca, Hypovirus 1, un nuevo Hypovirus identificado en Entouleca sp. procedentes de aguacate

Entouleca, Hypovirus 1, un nuevo Hypovirus identificado en Entouleca sp. procedentes de aguacate

Abstract

Con objeto de identificar elementos de biocontrol de hongos de raíz de aguacate, a partir de aislados del hongo Entoleuca sp., asociado a la raíz de aguacate, hemos detectado una serie de elementos dsRNA. Los dsRNA extraídos se han utilizado para su secuenciación masiva en plataforma Illumina y, tras posterior análisis BLASTn y BLASTx, han permitido detectar un a posible nueva especie vírica de la familia Hypoviridae. La secuencia borrador obtenida a partir de contigs de las secuencias masivas sirvió para, complementada con secuenciación convencional usando cebadores específicos, y determinación de los extremos 5’ y 3’ mediante el método de ligación de dsRNA con primer loop, obtener la secuencia completa de una nueva especie viral. El genoma de este virus, para el que proponemos el nombre de Entoleuca hypovirus 1 (EnHV-1, aislado E97-14), consta de 14.946 nts. Contiene un extremo 5’ no codificante de 945 nts, seguido de dos ORF contiguos de 1.770 y 10.920 nts. La proteína que aparentemente codifica el ORF A no tiene homología con proteínas conocidas ni tampoco ningún dominio reconocible. EL ORF B codifica para una proteína de alto peso molecular, probablemente una poliproteína con funciones de replicasa e incluye un dominio homólogo a helicasas y una región con homología a polimerasas de RNA. A continuación se encuentra un extremo 3’ no codificante de 1.279 nts, seguido de un poly-A. Además se ha secuenciado completamente el genoma de otro aislado de EnMV-1 (E112-4) procedente también de Entoleuca, lo que ha permitido el diseño de cebadores específicos para detectar el virus. Mediante RT -PCR empleando estos cebadores hemos podido identificar EnMV-1 en otros aislados de Entoleuca sp. y también en Rosellinia necatrix procedentes de aguacates cultivados en la costa subtropical. Actualmente estamos investigando los efectos de EnMV en la patogenicidad y Rosellinia en aguacate.
