Marcaje de bifidobacterias mediante una proteína verde fluorescente funcional en ausencia de oxígeno

Abstract

Actualmente contamos con información limitada acerca de la interacción de las bifidobacterias con otros miembros de la microbiota intestinal, así como sobre los mecanismos moleculares responsables de sus beneficios para la salud. Existe por tanto, la necesidad de identificar y monitorizar el comportamiento de las bifidobacterias de interés, tanto durante el proceso fermentativo del alimento, como en el tracto gastrointestinal. Esta tarea es especialmente complicada en el caso de las bifidobacterias debido a sus requerimientos metabólicos y su resistencia a la modificación genética. La GFP ha sido una herramienta molecular muy explotada para marcar y detectar bacterias en tiempo real dentro de ecosistemas complejos. Sin embargo, el oxígeno es indispensable para la formación de los cromóforos fluorescentes, y ha dificultado su uso en ambientes anaerobios. En el presente trabajo se ha desarrollado un vector (pNZ:Tu-GFPana) para el marcaje de bifidobacterias basado en el plásmido pNZ8048, que alberga el factor de elongación Tu de Bifidobacterium longum y una proteína verde fluorescente que contiene un cofactor basado en el flavin mononucleótido, el cual es fluorescente tanto en condiciones aerobias como anaerobias.
