[ES]: Aunque hay varios estudios que destacan la activación de la vía PI3K/Akt en mieloma, hay menos información sobre el estado y el papel de la vía mTOR. Aquí, exploraremos, utilizando técnicas moleculares, el estado de la vía mTOR en las células de mieloma primarias y líneas celulares. También emplearemos la interferencia de ARN de estas dos ramas de la vía mTOR para explorar el papel de estas dos vías en las células del mieloma. Trataremos de proporcionar evidencia sobre la importancia de la vía mTOR en el desarrollo de mieloma. Otra parte de este estudio es identificar el mecanismo de resistencia de las células del mieloma a TRAIL. La eficacia anti-mieloma de TRAIL se ha demostrado previamente. Sin embargo, la resistencia a TRAIL es un problema común para esta terapia dirigida no sólo en el mieloma, sino también en otros tipos de cáncer. Trataremos de demostrar los eventos moleculares involucrados en la resistencia de las células del mieloma a TRAIL a través de estudios sobre el modelo de cultivo in vitro de resistencia. [EN]: Although several studies highlighting the activation of the PI3K/Akt pathway in myeloma, there is less information on the status and role of the mTOR pathway. Here, we explore, using molecular techniques, the state of the mTOR pathway in primary myeloma cells and cell lines. We will also use RNA interference of these two branches of the mTOR pathway to explore the role of these two pathways in myeloma cells. Attempt to provide evidence of the importance of the mTOR pathway in the development of multiple myeloma. Another part of this study is to identify the resistance mechanism of myeloma cells with TRAIL. The anti-myeloma efficacy of TRAIL has been shown previously. However, TRAIL resistance is a common problem for this therapy directed not only in myeloma, but also in other types of cancer. Try to demonstrate the molecular events involved in the resistance of myeloma cells to TRAIL through studies in vitro culture model of resistance. These studies may facilitate the clinical use of TRAIL.
