Abstract

[ES] La selección in vitro es una herramienta utilizada tanto en los programas de mejora para disminuir el tiempo empleado en la obtención de una variedad comercial, así como en investigación para la selección de nuevas combinaciones génicas que implementen una característica fenotípica deseada. Los cultivos de cebada sufren diferentes estreses, bióticos y abióticos, que causan grandes pérdidas en el rendimiento de las cosechas. Entre ellos están la toxicidad del exceso de boro en suelo y la fusariosis de la espiga, una infección causada por hongos del género Fusarium. En este trabajo se desarrollaron dos métodos de selección in vitro. Uno para cribar individuos resistentes a estrés por boro y el otro para estrés por Fusarium. Se utilizó como material vegetal embriones cigóticos procedentes de plantas doble haploides. En el caso de la toxicidad al boro se aplicó directamente este elemento en el medio de selección. En el caso de la fusariosis y debido a que la resistencia a esta enfermedad presenta varios componentes, se estudió en concreto la resistencia a la acumulación de deoxinivalenol (DON). El DON es una de las toxinas producidas por Fusarium, es un factor de virulencia y fue la sustancia utilizada en el medio de selección para la realización del cribado.

[EN] In vitro selection is one techniquel used in breeding programs to reduce the time to get a commercial variety and in investigation to obtain new genic combinations that improve a desired phenotype. There are many stress, biotic and abiotic, that affect the yield of barley. Among them, we find the excess of soil boron toxicity and Fusarium Head Blight (FHB), a fungus infection caused by genus Fusarium. We developed two methods of in vitro selection in this work. One method was for the screening of plants resistant to boron stress and the other for plants resistant to scab. The plant material used were zygotic embryos recovered from double haploid plants. For boron toxicity, boron was applied directly into the selection media. For FHB, as it is a very complex disease, we studied the component of resistance for deoxinivalenol (DON) accumulation. DON is one of the toxins produced by Fusarium, as well as a virulence factor, and was the substance used in the selection media for carrying out the sreening
