Caracterización y análisis funcional de los genes homólogos de pttg1/securina, pttg2 y pttg3

Abstract

La securina humana, también conocida como PTTG1 (del inglés, human pituitary tumor-transforming gene 1), juega un papel esencial en la regulación del ciclo celular. Recientemente se han identificado dos genes homólogos de pttg1, pttg2 y pttg3, aunque sus funciones fisiológicas aún no se han descrito. En este estudio se ha llevado a cabo la caracterización detallada de estos genes homólogos de pttg1. Mediante PCR en tiempo real utilizando sondas específicas, hemos demostrado que Pttg2 se expresa a muy bajo nivel mientras que Pttg3 es prácticamente indetectable. Por tanto, nuestro trabajo se ha centrado fundamentalmente en el estudio de PTTG2. A lo largo de este análisis hemos demostrado que, a diferencia de PTTG1, PTTG2 carece de actividad transactivadora, así como de la capacidad para interaccionar con separasa y por tanto de función securina. Para estudiar el papel biológico de PTTG2, inhibimos su expresión mediante el uso de un short hairpin RNA (shRNA) contra dicha proteína. La bajada de los niveles de Pttg2 en la célula provocaba alteraciones en la morfología celular compatibles con un defecto en la capacidad de adhesión. Además, estas células carentes de Pttg2 morían por apoptosis dependiente de p53 y p21. Mediante la tecnología de microarrays, obtuvimos un perfil de expresión génica de células deficientes en Pttg2 respecto a células wild type. En este análisis observamos que la ausencia de Pttg2 provocaba una disminución de los niveles de E-cadherina y un aumento en los niveles de vimentina. Ambos efectos constituyen marcadores del fenómeno de EMT (Epithelial mesenchymal transition). La observación del comportamiento aberrante de las células carentes de Pttg2, reveló la contribución de dicha proteína al establecimiento de la adhesión celular así como al proceso de EMT y con ello posiblemente al fenómeno de invasión celular.
