Glutatión Peroxidasas y Tiorredoxinas en la Leguminosa Modelo Lotus japonicus

Abstract

Objetivos: 1. Determinar la función de las glutatión peroxidasas en los nódulos de leguminosas. 1.1. Caracterizar bioquímicamente las dos glutatión peroxidasas (LjGpx1 y LjGpx3) más abundantes en los nódulos de Lotus. 1.2. Determinar posibles modificaciones postraduccionales e investigar su efecto en la actividad biológica. 1.3. Estudiar la implicación de LjGpx1 y LjGpx3 en la respuesta al estrés abiótico y en mecanismos de señalización mediados por fitohormonas y óxido nítrico. 1.4. Producir anticuerpos altamente específicos frente a LjGpx1 y LjGpx3. 1.5. Localizar LjGpx1 y LjGpx3 en las células y tejidos de los nódulos utilizando diferentes aproximaciones experimentales. 2. Estudiar la familia de las tiorredoxinas en Lotus y determinar la función de la isoforma mitocondrial Trxo en el establecimiento y funcionamiento de la simbiosis. 2.1. Identificar y caracterizar los genes que codifican tiorredoxinas. 2.2. Determinar las dianas moleculares de tiorredoxinas de nódulos. 2.3. Producir Trxo (isoforma mitocondrial) recombinante. 2.4. Determinar la función de Trxo en las leguminosas mediante el uso de plantas transgénicas (RNAi) y mutantes knockout deficientes en Trxo.
