Abstract

Mediante la técnica de registro de corrientes iónicas macroscópicas que se puso a punto en el laboratorio se estudiaron las propiedades de la conductancia de potasio independiente de calcio de las membranas celulares de las celulascromafines continuando la descripción iniciada por A. Marty y E. Nehersen 1985 a nivel microscópico. Las conclusiones obtenidas en este trabajo fueron: 1. Las corrientes macroscópicas de potasio en la célula cromafin tienen un componente independiente de calcio que es semejante a la corriente de potasio que atraviesa el canal rectificador retrasado clásico por: 1) la forma de su relación intensidad-voltai. 2) su bloqueo por tea 3) su cinética de activación tipo n elevado a 4 y 4) el carácter óhmico de los canales iónicos abiertos que contribuyen a esta conductancia. 2. La activación de la conductancia de la membrana correspondiente a la corriente de potasio independiente de calcio dependen del potencial de membrana en forma similar a la correspondiente al rectificador retrasado clásico. Sin embargo esta dependencia es menos acusada (con una carga de compuerta equivalente de z=2e) y además el potencial de membrana al que se alcanza el 50 % de la activación es más despolarizado. 3) La i sobre k en la célula cromafin disminuye lentamente alcanzando un valor estacionario no nulo en unos 20 s aproximadamente. Esta disminución de la corriente se debe a una inactivación de la conductancia al potasio no dependiente de calcio y no a un fenómeno de acumulación. 4) El proceso de inactivación de la i sobre k no es una exponencial simple. La constante de inactivación más lenta depende del potencial de membrana haciéndose más rápida al aumentar este. 5) Pulsos despolarizantes repetidos causan inactivación de i sobre k. 6) La inactivación depende del potencial de membrana y su recuperación es lenta.
