Regulación de la longevidad por la sulfatasa sul-2 en Caenorhabditis elegans

Abstract

Los objetivos principales de esta Tesis Doctoral han sido identificar y caracterizar un nuevo gen que regula longevidad en el organismo modelo Caenorhabditis elegans. Previamente, en nuestro grupo, para descubrir nuevos loci implicados en envejecimiento en este nematodo se realizó una mutagénesis y búsqueda de nuevos mutantes longevos, entre los aislados se encontraba liv-7(pv17) que era longevo y carecía de otros fenotipos visibles, por lo que decidimos que podía ser muy interesante su identificación y caracterización. La mutación pv17 resultó ser un alelo del gen sul-2, homólogo a sulfatasa esteroidea, la encargada de quitar el sulfato de las hormonas esteroideas sulfatadas y de esta forma regular su actividad. Además de este alelo, que parece tener una reducción de función, hemos comprobado el fenotipo de la deleción de sul-2(gk187) y un inhibidor de hormonas esteroideas, el STX64 usado para el tratamiento del cáncer, y en ambos casos hemos comprobado que presenta fenotipos similares al alelo pv17.
