Organización de los nucleosomas y regulación genómica en Schizosaccharomyces pombe

Abstract

El genoma de los organismos eucariotas, está organizado en cromatina, una estructura altamente ordenada en la cual el DNA se empaqueta interaccionando con un gran número de proteínas de las que las histonas H2A, H2B, H3 y H4 son los componentes principales (Muñoz- Centeno et al., 2012). Éstas forman un octámero que contiene dos copias de cada una de ellas y alrededor del cual se enrollan 1.7 veces 147 pb de DNA que constituyen el nucleosoma, la unidad estructural básica de la cromatina (Richmond & Davey, 2003). Las primeras interacciones intermoleculares entre las histonas y el DNA fueron determinadas con una resolución de 2.8 Å mediante cristalografía de rayos X de un nucleosoma ensamblado a partir de sus componentes purificados (Luger et al., 1997). Un DNA espaciador de longitud variable entre especies y entre tipos celulares, conecta un nucleosoma con el siguiente. Además, la mayor parte de los eucariotas tienen una molécula de histona H1 unida al DNA espaciador y al nucleosoma en el punto donde el DNA sale y entra del octámero (Harshman et al., 2013). Este empaquetamiento del DNA en nucleosomas contribuye a la compactación del DNA genómico en estructuras de cromatina de orden superior.
