Abstract

[ES] Los nucleosomas facilitan el empaquetamiento del genoma en el n?cleo eucari?tico y modulan el acceso de los reguladores al ADN. Una descripci?n detallada de la organizaci?n y remodelamiento de la cromatina bajo diferentes condiciones fisiol?gicas y en especies distintas es fundamental para entender su contribuci?n a la regulaci?n y evoluci?n de los genomas. Para determinar la din?mica de la cromatina bajo programas transcripcionales diferentes hemos generado mapas de nucleosomas de alta resoluci?n en Schizosaccharomyces pombe. Nuestros resultados muestran que el 98,5% del genoma permanece pr?cticamente invariable durante mitosis y meiosis, mientras que el remodelamiento se limita a la exclusi?n de 1100 nucleosomas aproximadamente localizados en los promotores de un subconjunto de genes expresados diferencialmente durante meiosis. Estas regiones libres de nucleosomas (NDRs) inducibles y las que est?n presentes de forma constitutiva en el genoma solapan de forma precisa con grupos de sitios de uni?n para factores de transcripci?n (TF) espec?ficos de meiosis y de diferentes clases funcionales de genes respectivamente. La deleci?n individual de dos TFs afecta solamente a una peque?a fracci?n de todas las NDRs a las que se unen in vivo, indicando que los factores de transcripci?n contribuyen colectivamente al mantenimiento de las NDRs. Adem?s, el an?lisis de otras dos especies del mismo g?nero, S. octosporus y S. japonicus, muestra que la organizaci?n nucleosomal de regiones transcritas e interg?nicas y los motivos reguladores encontrados en las NDRs gen?micas est?n muy conservados en las levaduras de fisi?n, siendo el grado de conservaci?n mayor entre S. pombe y S. octosporus, las dos especies m?s pr?ximas evolutivamente. Nuestros resultados en tres especies diferentes revelan un patr?n de nucleosomas estricto y constante que favorece la concentraci?n de los reguladores en las NDRs. El an?lisis de los grupos de motivos reguladores en esta fracci?n discreta del genoma facilitar? la definici?n de las redes regulatorias eucari?ticas.
