Obtención de extractos enriquecidos en ácido rosmarínico a partir de mejorana

Abstract

La mejorana (Origanum majorana L.) es una hierba perteneciente a la familia Lamiaceae, empleada tradicionalmente para el tratamiento de síntomas gastrointestinales, enfermedades bronquiales y congestión nasal, entre otras. Debido a sus propiedades, los compuestos bioactivos presentes en la mejorana han sido estudiados, concluyéndose en aceites esenciales, compuestos fenólicos y labiataetaninos, destacando el ácido rosmarínico (AR) al cual se le confieren actividades biológicas como antiviral, antibacterial, anti-inflamatorio y antioxidante. Tradicionalmente la extracción de estos compuestos ha sido mediante extracciones sólido-líquido convencionales, concluyendo que las mezclas de agua-metanol han sido las más efectivas. El objetivo de este estudio fue determinar las condiciones óptimas de extracción de AR mediante fluidos presurizados (PLE) a partir de mejorana. Para ello se realizaron extracciones a diferentes condiciones de temperatura (100ºC y 150ºC) y concentraciones de metanol-agua (30% metanol 50% metanol, 70% metanol). La composición de AR en cada uno de los extractos fue determinada a partir de HPLC-DAD. Los resultados obtenidos muestran que el rendimiento de extracción de AR resultó diferente para las dos temperaturas estudiadas, siendo superior a 100ºC. Así mismo, se observaron diferencias para cada concentración de disolvente, siendo las condiciones de 100ºC y 50% metanol-agua las que presentaron mejores resultados (12,7 mg AR/g extracto). Los valores obtenidos a estas concentraciones resultaron similares a los reportados por otros autores. Futuros estudios consistirán en la obtención de concentrados de AR mediante técnicas de purificación con resinas XAD-7HP y ultrafiltración con membranas, para finalmente determinar su capacidad antioxidante mediante el método de ORAC y su capacidad anti-inflamatoria de en modelos celulares THP-1.
