Abstract

La importancia de los polifenoles y en concreto de los flavan-3-oles en la salud está firmemente apoyada por distintos estudios científicos. La bioactividad de estos compuestos depende de su biodisponibilidad, absorción y metabolismo en el individuo. Así, la microbiota intestinal juega un papel esencial al transformar estos compuestos fenólicos en metabolitos con distinta bioactividad y significación fisiológica. La degradación bacteriana de los polifenoles se ve asimismo afectada por los efectos antibacterianos inherentes a estas moléculas. Estudios previos in vitro realizados con incubaciones estáticas de cepas bacterianas puras han mostrado la capacidad del género Lactobacillus para crecer en presencia de extractos polifenólicos ricos en flavan-3-oles. L. plantarum IFPL935 mostró además el potencial enzimático necesario para metabolizar ciertos monómeros de flavan-3-oles y derivados galoilados. Esta cepa destaca por la actividad enzimática, sólo descrita en ciertas bacterias intestinales, que inicia la rotura del anillo heterocíclico de monómeros de flavan-3-oles produciendo el difenilpropan-2-ol correspondiente. En este trabajo se ha evaluado el papel de L. plantarum IFPL935, como integrante de la microbiota intestinal, en el metabolismo de flavan-3-oles. Así, se han realizado incubaciones in vitro de IFPL935 y un extracto polifenólico de vino, junto con una microbiota representativa de las distintas regiones del colon desarrolladas en un simulador de la microbiota intestinal humana (SHIME). Los metabolitos polifenólicos se determinaron por cromatografía líquida (UPLC). Los resultados han mostrado que L. plantarum IFPL935 aumenta significativamente la producción de difenilpropan-2-ol, confirmando que aún en presencia de una microbiota colónica compleja esta cepa puede iniciar el metabolismo de los flavan-3-oles hacia metabolitos con mayor significación biológica.
