Apertura de sulfamidatos cíclicos con nucleófilos nitrogenados

Abstract

Esta tesis doctoral se centra en el estudio de la reacción de apertura de sulfamidatos cíclicos derivados de alfa-metilisoserina frente a nucleófilos nitrogenados, ampliando así los trabajos previos realizados en nuestro grupo sobre estos sulfamidatos cíclicos con nucleófilos oxigenados y azufrados. En primer lugar se abordó el estudio de reactividad del sulfamidato derivado de alfa-metilisoserina frente a diferentes aminas, observándose distinta reactividad según la basicidad de las mismas. Se optimizó la reacción de apertura nucleófila para arilaminas primarias y alfa-aminoésteres. Posteriormente, se continuó este estudio frente a diferentes aminas aromáticas, como imidazoles y pirazol, obteniendo los productos de apertura del sulfamidato con buenos rendimientos. Por último, se completó el estudio utilizando como nucleófilos diversas piridinas y N-alquilimidazoles, llegando a los correspondientes productos de apertura con excelentes rendimientos. Se ha demostrado que la reacción de apertura nucleófila del sulfamidato transcurre con total inversión de configuración del centro cuaternario. Esta metodología ha permitido el acceso, de forma enantioméricamente pura, a diferentes compuestos de gran interés en síntesis orgánica como son cuatro nuevos ß-aminoácidos alfa,alfa-disustituidos, dos de ellos cargados, dos nuevos bis-aminoácidos análogos a la histidinoalanina y una nueva piperazinona. Por último, se ha comenzado el estudio de la síntesis de carbenos quirales de plata a partir de las sales de imidazolio obtenidas de las reacciones de apertura de sulfamidatos con N-metilimidazol.
