Abstract

En esta tesis doctoral se ha desarrollado una nueva estrategia sintética que permite abordar la síntesis de alfa-metilaminoácidos enantioméricamente puros por alquilaci]on diastereoselectiva de 2- cianoésteres quirales derivados de (1S,2R,4R)-10-diciclohexilsulfamoílisoborneol y posterior manipulaci]on de los grupos funcionales presentes en el producto de alquilaci]on para así generar los grupos amino y ácido del alfa-metilaminoácido final. Del estudio realizado del proceso de alquilaci]on puede concluirse que: - Es necesaria la utilizaci]on de halogenuros reactivos para que el proceso transcurra con elevados rendimientos y diastereoselectividades. - Si el halogenuro es lo suficientemente reactivo puede utilizarse como base carbonato potásico en lugar de diisopropilamiduro de litio, obteniéndose el producto de alquilaci]on con rendimientos elevados y observándose en algunos casos un ligero descenso de la diastereoselectividad del proceso. - La utilizaci]on de triflatos como electr]ofilos no parece aconsejable en nuestro caso a la vista de los rendimientos y diastereoselectividades observadas con su utilizaci]on. El acceso a un aminoácido de una determinada configuraci]on puede abordarse de tres modos diferentes: - Eligiendo el cianoéster de partida y el agente de alquilaci]on. - Efectuando procesos de degradaci]on alternativa a partir de un precursor común. - Utilizando ambos enanti]omeros del auxiliar quiral. De este modo se han sintetizado los siguientes alfa-metilaminoácidos: (R) y (S)-alfa-metilfenilalanina, (S)-alfa-metiltript]ofano, (R)-alfa- metildifenilalanina, (R)- isovalina, (R) y (S)-alfa-metilvalina, ácido (S)-amino-2-metilpentanoico, ácido (S)-2-amino-2,5-dimetil- hexanoico, (S)-alfa-metileucina, (R)-alfa- metilfenilglicina, ácido (S)-alfa-metilaspártico y (S)-alfa-metilserina. Por último, utilizando la sistemática adecuada se ha logrado introducir un aminoácido muy impedido estéricamente en cadenas peptídicas llegando al nivel de pentapéptido. En definitiva, se ha desarrollado una nueva metodología, sencilla y versátil que permite, a partir de productos fácilmente accesibles y a través de procesos sencillos, la obtenci]on de un gran número de alfa-metilaminoácidos enantioméricamente puros.
