Eficacia de la endolisina LysH5 en la eliminación de biofilms formados por Staphylococcus

Abstract

Los “biofilms” (biopelículas) producidos por algunas especies pertenecientes al género Staphylococcus , en especial Staphylococcus aureus y Staphylococcus epidermidis , constituyen un serio problema en los sectores alimentario y hospitalario . En estos ecosistemas bacterianos los microorganismos quedan protegidos por una matriz extracelular que impide la penetración de antibióticos y desinfectantes, dificultando su eliminación . Los bacteriófagos y en concreto las proteínas líticas derivadas de ellos (endolisinas), son alternativas prometedoras para combatir este problema . El objetivo de este trabajo ha sido estudiar la eficacia de la endolisina LysH 5 , codificada por el fago vB_SauSIPLA 88 de S . aureus , para eliminar biofilms formados por S . aureus y por S . epidermidis . Se determinó además la sensibilidad de las células supervivientes a esta proteína y su capacidad para volver a formar biofilms . Finalmente, se comprobó la ausencia de bacterias “ persisters ” (células en estado durmiente, que resisten la acción de antibióticos, y que están relacionadas directamente con el fallo de los tratamientos antibióticos en terapia clínica ) tras el tratamiento de cultivos con LysH 5 , así como la capacidad de esta proteína para eliminar este tipo de bacterias.
