Los segmentos K de la dehidrina DHN1a de uva de mesa son esenciales para conferir actividad crioprotectora, antifúngica y de protección frente a la deshidratación in vitro.

Abstract

La uva de mesa tiene limitada su conservación a baja temperatura debido a su alta susceptibilidad a la podredumbre por hongos y su sensibilidad a la pérdida de agua después de la cosecha. Trabajos previos de nuestro grupo han mostrado la eficacia de un pre-tratamiento de 3 días con alta concentraciones de CO2 a 0ºC manteniendo la calidad de la uva de mesa. Las dehidrinas se acumulan en respuesta a estreses ambientales que tienen un componente de pérdida de agua intracelular, como son bajas temperaturas y sequía. Mediante RT-PCR hemos aislado, a partir de uvas de mesa, un cDNA que codifica una dehidrina tipo YSK2 (VvcDHN1a), que presentó un “splicing alternativo” que consistió en la retención de un intrón de 84 pb dando como resultado una proteína truncada del tipo YS. Tanto el almacenamiento a baja temperatura como el pre-tratamiento gaseoso indujeron transcritos con y sin “splicing alternativo” aunque con diferentes patrones de acumulación en el tiempo. La falta de los segmentos K, característicos de esta familia, podría afectar a la funcionalidad de la forma truncada de DHN1a. Con el objetivo de confirmar esta hipótesis se ha llevado a cabo la sobreexpresión de la proteína YSK2 y de la forma truncada YS en Escherichia coli. Se ha estudiado el posible papel protector de actividades enzimáticas de ambas proteínas recombinantes frente a estreses abióticos como la congelación y la deshidratación, así como su posible función antifúngica. Los resultados han mostrado que la proteína madura YSK2 confiere protección in vitro frente a la congelación, la deshidratación así como cierta actividad antifúngica frente a Botrytis cinerea. En el caso de la proteína truncada no se han observado estos efectos protectores concluyéndose que las funciones de protección otorgadas a VvcDHN1a podrían estar directamente relacionadas con la presencia de sus segmentos K.
