Abstract

En la presente Tesis Doctoral se ha estudiado el aislamiento,caracterizacion y estudio de las propiedades funcionales y biologicas del Caseinmacropeptido aislado a partir de concentrados de suero y suero de leche de oveja, vaca y cabra. En primer lugar se desarrolló un método de aislamiento en planta piloto, tras el cual se llevó a cabo una validación del mismo mediante el estudio del rendimiento y grado de pureza del producto final. Los resultados del presente demuestran que el metodo reduce el número de pasos, presenta un alto grado de pureza y rendimiento, permite la utilizacion de sueros de queseria enorme problema medioambiental y no utiliza productos tóxicos como el TCA, muy utilizados en otros metodos de asilamiento. Por otro lado se llevó a cabo un estudio analitico de todos los CMP aislados con la finalidad de compararlos entre ellos y encontrar diferencias intra e interespecificas. Se estudio su contenido en nitrogeno total, nitrogeno no proteico, lactosa, acido lactico, aminoacidos, Nana, minerales, asi como un estudio mediante RP-HPLC y SDS-PAGE. Los resultados demuestran que no hay diferencias significativas entre ellos, confirmandose una de sus propiedades mas importantes como es la ausencia de aminoácidos aromaticos y un perfil cromatográfico similar en oveja y cabra y diferente en vaca. Por último se llevo a cabo el estudio de las propiedades funcionales, estudio que pemite el análisis de su solubilidad, capacidad gelificante, capacidad espumante, viscosidad y termorresistencia. Para ello se desarrolló un RSM (metodologia de superficie respuesta) que permitia minimizar el número de ensayos en unas determinada condiciones previamente establecidas y que nos ayudan a modelizar y predecir el comportamiento en todo el intervalo. Los resultados demuestran que las diferencias entre los CMP y el WPC son muy significativas, seguramente debido a la diferencia estructura de sus componentes.
