Abstract

La manipulación genética se ha convertido en un método corriente de análisis y caracterización de genes eucarióticos y sus productos. Los hongos constituyen un grupo de organismos los cuales han demostrado ser muy útiles para la elucidación de algunos principios genéticos, y Podospora anserina ha sido uno de ellos. Sin embargo, la mayoría de los mutantes obtenidos hasta el presente en esta especie lo han sido de tipo morfoldgico, lo cual no facilta mucho la selección de los productos obtenidos tras la manipulacidn. La posesión de un plásmido mitocondrial por parte de P. anserina hace que sea aún más interesante la genética de este hongo, abriendo un nuevo camino hacia los procesos de transformación de células eucarióticas y consecución de un posible vehículo de clonación para hongos filamentosos. En el presente trabajo se describen los procesos para la obtención de cepas resistentes a diferentes antifúngicos inhibidores del crecimiento, con las cuales sea posible la obtención de una serie de marcadores claros, sencillos y fácilmente utilizables para procesos de manipulación genética.

Genetic manipulation has become a common method for the analysis and characterization of eukaryotic genes and their products. Fungi are a group of organisms which have proven very useful in elucidating certain genetic principles. and Podospora anserina has been one of them. However, most of the obtained mutants hitherto in this species are morphological, and they do not help very much in the selection of the manipulation products. As P. anserina has a mitochondrial plasmid, the genetics of this fungus becames more interesting, and opens a new way for the transformation processes in eukaryotic cells and construction of a possible cloning vehicle in filamentous fungi. In this work are described the processes for the isolation of resistant strains to different antifungic inhibitors, which can be useful for the obtention of easy, simple and clear markers useful for the genetic manipulation processes.
