Química y electroquímica de derivados del cloral en la síntesis de triazinas, oxadiazolinas, piridazinas y pirazolonas

Abstract

Se han desarrollado nuevos y eficientes métodos de síntesis de compuestos heterocíclicos nitrogenados, tales como triazinas, oxadiazolinas, piridazinas y pirazolonas. También se han efectuado estudios computacionales complementarios basados en la teoría del funcional de la densidad (DFT). Las reacciones entre cloral y benzamidas dieron lugar a cloralbenzamidas, las cuales fueron transformadas en cloruros de N-(1,2,2,2-tetracloroetil)bencimidoílo. Estos compuestos reaccionaron con guanidina proporcionando 2-amino-4-aril-6-diclorometil-1,3,5-triazinas. Estos mismos cloruros de imidoílo se utilizaron como intermedios en la síntesis de 4-aryl-2-hydroxy-1,3,5-triazinas a través de una secuencia de reacciones con amoniaco y trifosgeno. Las estructuras moleculares de 2-amino-6-diclorometil-4-fenil-1,3,5-triazina y 6-(4-clorobencil)-4-triclorometil-3,4-dihidro-1,3,5-triazin-2(1H)-ona fueron determinadas mediante cristalografía de rayos X. N-(2,2-diclorovinil)benzamidas, generadas electroquímicamente, fueron transformadas en cloruros de N-(2,2-diclorovinil)bencimidoílo, los cuales reaccionaron con hydroxilamina para dar 3-aril-5-diclorometil-1,2,4-oxadiazolinas. La estructura molecular de 5-diclometil-3-(4-fluorobencil)-1,2,4-oxadiazolina fue determinada por difracción de rayos X. Se describe, y se ha estudiado con la ayuda de cálculos DFT, un tipo de reagrupamiento previamente desconocido de 3-aril-5-diclorometil-2-pirazolinas a 3-arilpiridazinas, promovido por terc-butóxido potásico. El 4,4,4-triclorocrotonato de etilo reacciona con hidrato de hidrazina dando 5-triclorometil-3-pyrazolidona, cuyo tratamiento con alquilisocianatos proporciona 1-alquilaminocarbonil-5-triclorometill-3-pirazolidonas. La reducción electroquímica de estas sustancias seguida de reacción con terc-butóxido potásico condujo a nuevas 2-hidroxi-5-alquil-4,5-dihidroimidazo[1,5-b]pirazol-6-onas. La estructura molecular de 2-hidroxi-5-pentil-4,5-dihidroimidazo[1,5-b]pirazol-6-ona fue determinada mediante difracción de rayos X. Efficient new synthetic methods for several classes of nitrogen-containing heterocyclic compounds, including triazines, oxadiazolines, pyridazines and pyrazolones have been developed starting from chloral derivatives. Complementary theoretical studies based on density functional theory (DFT) have been carried out. Reactions between chloral and benzamides gave chloralbenzamides, which were transformed to N-(1,2,2,2-tetrachloroethyl)benzimidoyl chlorides. These reacted with guanidine providing previously unknown 2-amino-4-aryl-6-dichloromethyl-1,3,5-triazines. 4-Aryl-2-hydroxy-1,3,5-triazines were synthesized from the same imidoyl chloride intermediates by sequential reactions with ammonia and triphosgene. The molecular structures of 2-amino-6-dichloromethyl-4-phenyl-1,3,5-triazine and 6-(4-chlorobenzyl)-4-trichloromethyl-3,4-dihydro-1,3,5-triazin-2(1H)-one were determined by X-ray crystallography. Electrochemically generated N-(2,2-dichlorovinyl)benzamides were transformed to N-(2,2-dichlorovinyl)benzimidoyl chlorides, which reacted with hydroxylamine yielding 3-aryl-5-dichloromethyl-1,2,4-oxadiazolines. The X-ray molecular structure of 5-dichlomethyl-3-(4-fluorobenzyl)-1,2,4-oxadiazoline was determined. A hitherto unknown rearrangement of 3-aryl-5-dichloromethyl-2-pyrazolines to 3-aylpyridazines, promoted by potassium tert-butoxide, is described and studied with the aid of DFT calculations. Ethyl 4,4,4-trichlorocrotonate reacted with hydrazine hydrate to give 5-trichloromethyl-3-pyrazolidone, whose treatment with alkylisocyanates provided hitherto unknown 1-alkylaminocarbonyl-5-trichloromethyl-3-pyrazolidones. Electrochemical reductions of these substances followed by reaction with potassium tert-butoxide led to novel 2-hydroxy-5-alkyl-4,5-dihydroimidazo[1,5-b]pyrazol-6-ones. The X-ray molecular structure of 2-hydroxy-5-pentyl-4,5-dihydroimidazo[1,5-b]pyrazol-6-one was determined.
