Abstract

Free radicals are involved in many human diseases, including Alzheimer´s disease and Parkinson´s disease. The brain is thought to be especially vulnerable to oxidative damage, given its high oxygen consumption, the high levels of polyunsaturated fatty acids and nonregenerative neurons and rather low levels of antioxidant enzymes. In this work, hippocampal lipid peroxidation and catalase enzyme activity in the hippocampus and striatum of rats treated with the fungicide maneb (30 mg/kg) and the herbicide paraquat (10 mg/kg) were estimated by spectrophotometric methods. Neither lipid peroxidation nor the enzymatic activity in the hippocampus were affected in rats treated with either of the pesticides. On the other hand, inhibition of catalase activity was observed in the striatum of rats treated with either maneb or paraquat, indicating a weakening of the enzymatic defense system against free radicals. These findings suggest, preliminarily, a greater toxic action in dopaminergic regions. Understanding the biological effects of these chemicals is important in determining the roles these compounds may play in disease etiologies.

Os radicais livres estão envolvidos em um grande número de enfermidades do ser humano. O cérebro tem níveis baixos de enzimas antioxidantes e um conteúdo lípidico elevado, tornando-se muito susceptível ao ataque de espécies reativas de oxigênio. Neste trabalho avaliou-se a lipoperoxidação em hipocampo e a atividadeda enzima catalase em estriado e hipocampo de ratos tratados com o fungicida maneb (30 mg/kg) e o herbicidaparaquat (10 mg/kg). Não houve alteração nalipoperoxidação nem na atividade enzimática no hipocampo dos animais tratados com ambos os praguicidas, porém foi observada uma inibição da catalase no estriado dos ratos tratados com maneb e comparaquat. Com estes resultados pode-se sugerir, de forma preliminar, uma ação tóxica maior sobre centros dopaminérgicos. Estudos sobre a toxicidade destes compostos são essenciais na compreensão do papel destes praguicidas e dos radicais livres na etiologia das doenças.
