Abstract

A utilizão de microrganismos com o objectivo de avaliar a toxicidade de poluentes ambientais tem sido uma metodologia aceite devido à sua relativa precisão, rapidez e simplicidade. Os microrganismos têm diversos atributos que os tornam os seres vivos ideais para os bioensaios. As bactérias são fáceis de manusear e são sensíveis a muitos poluentes, respondendo muito rápido e reprodutivelmente a alterações no seu ambiente. Neste trabalho, com recurso a técnicas de delineamento experimental pretendeu-se avaliar o efeito dos factores ambientais: temperatura, pH e luz no crescimento da bactéria Vibrio fischeri, durante o teste da inibição do crecimento do Vibrio fischeri, bem como optimizar um meio de cultura bacteriano com o menor número de nutrientes orgânicos (baseado no meio de cultura descrito na norma deste teste) que permita manter válido o teste. Isto é, um meio de cultura em que a população bacteriana se multiplique 3 a 4 gerações, num período de incubação de 6 a 7 horas e que permita manter a sensibilidade de resposta aos poluentes.
