Mecanismos de ação antimicrobiana de óleos essenciais de plantas condimentares de família Lamiaceae

Abstract

No presente estudo procedeu-se à caracterização química e avaliação da atividade antimicrobiana de óleos essenciais de sete espécies de plantas aromáticas autóctones da região do Alentejo, nomeadamente: Calamintha nepeta, Lavandula stoechas subsp. luisieri, Lavandula pedunculata, Lavandula viridis, Mentha pulegium, Mentha spicata e Origanum vulgare subsp. virens. Os óleos essenciais em estudo apresentaram uma composição química diversificada, rica em monoterpenos oxigenados e revelaram um largo espetro de ação antimicrobiana, com capacidade inibitória do crescimento bacteriano de estirpes gram-positivas e gram-negativas. Observou-se também o efeito sinergético e/ou aditivo de misturas de dois óleos essenciais com elevada diminuição dos valores de concentração mínima inibitória. As misturas de óleos essenciais mais promissoras foram as de C. nepeta/L. pedunculata, C. nepeta/M. spicata e L. pedunculata/M. pulegium, as quais apenas não demonstraram efeito sinergético face a uma estirpe bacteriana. Os óleos essenciais estudados apresentaram alta toxicidade face a Artemia salina, com valores de CL50 compreendidos entre 35,0 e 213,6 mg/mL e baixa toxicidade quando administrados por via oral a murganhos (DL50 > 2000 mg/kg). O elevado potencial antimicrobiano dos óleos essenciais das plantas aromáticas condimentares em estudo sugere a sua potencial aplicação na indústria alimentar como preservantes de alimentos bem como nutracêuticos e o seu estudo na indústria farmacêutica, no sentido de avaliar a sua toxicidade para mamíferos e definir a dose terapêutica segura; Abstract: Mechanisms of Antimicrobial Action of Essential Oils of Condiment Plants of Lamiaceae Family In the present study the chemical characterization and evaluation of the antimicrobial activity of essential oils of seven species of autochthonous aromatic plants of the Alentejo region was performed, namely: Calamintha nepeta, Lavandula stoechas subsp. luisieri, Lavandula pedunculata, Lavandula viridis, Mentha pulegium, Mentha spicata and Origanum vulgare subsp. virens. Essential oils presented diverse chemical composition, rich in oxygenated monoterpenes and revealed a broad spectrum of antimicrobial action, with inhibitory capacity for bacterial growth of gram-positive and gram-negative strains. The synergistic and/or additive effect of mixtures of two essential essential oils was also observed, with a high decrease in the minimum inhibitory concentration values. The most promising essential oils mixtures were C. nepeta/L. pedunculata, C. nepeta/M. spicata and L. pedunculata/M. pulegium, which only showed no synergistic effect against one bacterial strain. Essential oils studied presented high toxicity to Artemia salina, with LC50 values between 35.0 and 213.6 mg/mL and low toxicity when administrated orally to mice (LD50 > 2000 mg/kg). The high antimicrobial potential of the essential oils of the spice aromatic plants under study suggests their potential application in the food industry as food preservatives as well as nutraceuticals and their study in the pharmaceutical industry, in order to assess their toxicity to mammals and to define the safe therapeutic dose
