Abstract

Alates esimestest vana DNA töödest on arheogenoomika teinud läbi suure arengu. Lisaks luudele ja mumifitseerunud/kuivanud pehmetele kudedele saab geneetilist informatsiooni sulgedest, koproliitidest, pinnasest, munakoortest jne. Kuna erinevatest bioarheoloogilistest materjalidest on DNA eraldamine erinev, on vaja DNA ekstraheerimise ja sekveneerimisraamatukogude loomise metoodikaid pidevalt täiustada, et väärtuslikku materjali maksimaalselt, kuid säästvalt kasutada. Käesolevas töös testiti kommertslike ning eelkõige kaasaegsete materjalide käsitsemiseks mõeldud ekstraheerimiskomplektide protokollide tingimusi erinevat päritolu bioarheoloogilisel materjalil ning leiti, et keskaegsetest nahaproovidest DNA ekstraheerimiseks valitud erinevad lüüsimisajad statistiliselt olulisi erinevusi ei andnud. Samas leiti, et keskaegse kuivanud koproliidiproovi koguse vähendamine tõstis tuntavalt DNA saagist ekstraktis. Genoomsete sekveneerimisraamatukogude loomise protokollide võrdlemisel leiti, et uus nn ühe tuubi protokoll kaasas pikemaid DNA fragmente, kuid rohkem PCR-i duplikaate, kui tavaprotokoll.
