Autoantikehade teke Aire-puudulikul rotil

Abstract

Käesolevas magistritöös uuriti I tüüpi interferoonide (IFN) vastaste autoantikehade teket ja nende mõju katselooma organismis. Selleks kasutati rotte, kellel oli välja lülitatud Aire (autoimmuunne regulaator) geen. Aire-puudulikel rottidel tekkisid IFN-α vastased antikehad enne sümptomite ilmumist, keskmiselt 4 kuu vanuselt. IgG kontsentratsiooni alusel normaliseeritud anti-IFN-α väärtused olid enamasti põrna kultuuris kõige kõrgemad, järgnesid tüümuse ja luuüdi rakkude supernatandid. Autoantikehadel oli interferoone neutraliseeriv efekt. Kõrge autoantikehade tiitri puhul olid muutunud T-raku alampopulatsioonide proportsioonid põrnas ja lümfisõlmedes. T follikulaarsete regulatoorsete rakkude populatsioon suurenes ning T follikulaarsete helperite populatsioon langes võrreldes heterosügootsete rottidega. Sooritati ka geeniekspressiooni analüüs, milles leiti, et alla on reguleeritud I tüüpi IFN-ga seotud geenide ekspressioonid, mis viitab, et autoantikehad on bioloogiliselt aktiivsed lisaks in vitro-le ka in vivo
