Wfs1 -puuduliku roti suhkurdiabeedi fenotüübi kirjeldamine

Abstract

Wolframi sündroom (WS) on autosomaalne retsessiivne neurodegeneratiivne haigus, mida põhjustavad mutatsioonid WFS1 geenis. Sündroomi põhisümptomid on magediabeet, suhkurdiabeet, nägemisnärvi atroofia ja sensorineuraalne kurtus. Et uurida WS-i ja võimalikke WS-i vastaseid raviteraapiaid, on praegu disainitud eri laborites kolm Wfs1 -puudulikku hiiremudelit. Asjaolu, et nimetatud hiirtel pole seni selgelt näidatud suhkurdiabeedi kujunemist, ajendas Tartu Ülikooli teadlasi konstrueerima Wfs1 -mutantse roti. Käesolevas töös uuriti Wfs1 -mutantsel Wfs1 -ex5-KO232 rotil suhkurdiabeedi fenotüüpi ja selle tekkepõhjuseid. Leiti, et Wfs1 -puuduliku roti pankrease beeta-rakkudes on tõusnud ER-i stressitase, mistõttu beeta-rakud hävivad ja 11. elukuul tekib insuliinist sõltuv suhkurdiabeet. Seega võib Wfs1 -puudulik rott olla uus mudel suhkurdiabeedi ja võimalike WS-i vastaste ravimite uurimiseks.
