WFS1-puudulikkusega roti piklikaju neurodegeneratsiooni iseloomustus

Abstract

Wolframi sündroomi (WS) põhjustab WFS1 geeni bialleelne mutatsioon. WS-i iseloomustavad suhkurdiabeet, endokriinne väärtalitlus, optiline atroofia ning ajutüve progressiivne neurodegeneratsioon. Eelnevad uuringud on peamiselt keskendunud esimese sümptomina avalduva suhkurdiabeedi uurimisele ning sellest lähtuvalt on neurodegeneratsiooni uurimine jäänud tagaplaanile. Käesolevas töös uuriti WFS1-puudulikkusega roti piklikaju neurodegeneratsiooni. Uuringus nähti WFS1-puudulikel rottidel piklikaju ruumala vähenemist, ajast sõltuvat neuronaalset ER-i stressi ning WFS1 valgu ekspressiooni vähenemist oliivtuumade tasandis. Lisaks näidati esmakordselt WFS1 lokaliseerumist FOXP2 positiivsetes oliivtuuma neuronites. Töö tulemustest võib järeldada, et WFS1-puudulikel rottidel esineb WS-i patsientidega sarnane piklikaju neurodegeneratsiooni muster.
