Papel del factor de transcripción Myc en el desarrollo del páncreas postnatal

Abstract

El desarrollo del páncreas está determinado por factores intrínsecos, como son el número de células progenitoras tempranas y por señales extrínsecas que concomitan con la diferenciación de los tipos de células endocrinas, exocrinas y ductales de una población única de precursores pancreáticos. El factor de transcripción Myc ha sido estudiado ampliamente como protooncogén, aunque en células humanas normales cumple un importante papel en el desarrollo embrionario de diversos tejidos entre ellos el páncreas, siendo un regulador de la maduración y mantenimiento de las células acinares de dicho órgano. La herramienta iFlpMosaics v2.0 introducida en ratones y desarrollada por el Grupo de Genética Molecular de la Angiogénesis del CNIC, representa un sistema inducible de mosaico genético funcional que permite inducir en el mismo tejido, células mutantes y wild-type y estudiar la competición entre ellas en el mismo entorno celular. Es por ello, que el objetivo de este estudio fue caracterizar la herramienta iFlpMosaics v2.0 en tejido pancreático y estudiar la actividad del factor de transcripción Myc en el desarrollo del páncreas postnatal. Mediante estudios de microscopia confocal multiespectral se pudo comprobar el funcionamiento de la herramienta iFlpMosaics recientemente desarrollada tanto en tejido de páncreas endocrino como exocrino. Además, el uso de esta herramienta ha permitido demostrar que células deficientes en el factor de transcripción Myc contribuyen mínimamente al desarrollo postnatal del páncreas murino.
