Abstract

La eritropoyetina es una glicoproteína acídica compuesta por 165 aminoácidos, sintetizada mayoritariamente en los riñones y en menor medida en la médula ósea, el bazo y el hígado. Gracias al desarrollo de la ingeniería genética, cada vez es más habitual la presencia de esta proteína tanto en el ámbito sanitario, como en el deportivo siendo utilizada como método de dopaje. En el presente trabajo, se estudia la estructura, biosíntesis, función de la proteína, sus diferentes indicaciones clínicas, y los efectos adversos que conlleva el abuso de esta hormona, dentro de los cuales se encuentra el cáncer. Se exponen las principales estrategias del dopaje con esta hormona, basadas principalmente en el dopaje genético y en la administración de proteínas recombinantes. Se indican los métodos utilizados en el dopaje genético y se explica el complejo proceso de obtención de las proteínas recombinantes, sus características principales y el uso en la mejora del rendimiento deportivo. En relación con el deporte, se detallan los diferentes métodos de detección de esta proteína en los controles antidopaje (los cuales son cada vez más exhaustivos) y también se exponen los derivados de la eritropoyetina que tienen un efecto más duradero en el organismo y son más difíciles de ser detectada. De esta manera se trata de acercar al lector al mundo del dopaje, profundizando en el conocimiento de su base molecular e informando sobre los avances que se están llevando a cabo para conseguir una competición más pura y limpia.

Erythropoietin is an acidic glycoprotein composed by 165 amino-acids, mainly synthesized in kidneys and in minor proportion in bone marrow, spleen and liver. Due to the development of genetic engineering it is increasingly common the presence of this protein in the scope health and in sport being used like a doping method. In the present report, the structure, the biosynthesis, the functions, the clinical indications and the adverse effects that are produced by the abuse of this protein, as cancer, are included. The main strategies of EPO doping are exposed, which are mainly based on gene doping and the administration of recombinant proteins. The most used methods in the gene therapy are indicated and the difficult process to obtain recombinant proteins, their main features and use in performance enhecement are explained. In relation to sport, the different detection methods of this protein in the anti-doping controls (which are more exhaustive now) and also erythropoietin derivatives las an option of doping substance are exposed, which have a long-lived effect in the organism and are more difficult to detect. In this manner, the work hope close lector to the doping world, going more deeply into the knowledge of their molecular basis, and give information about the advances that are being done to have a competition more pure and clean in every moment.
